The seminar will cover two separate projects, both related
to translational control in mammalian cells.
1. Traslational control during mitosis.
Global protein synthesis is transiently inhibited during
mitosis while translation of specific mRNAs is resistant to
down-regulation.
Our recent data indicates that the translational block is
downstream of the (currently accepted) initiation level.
The new concept of translational regulation at the
elongation level raises new challenging questions in the
field.
2. Translational control and brain development /
neurodegeneration
A point mutation in translation initiation eIF2B leads to a
devastating neurodegenerative genetic disease.
Surprisingly, although eIF2B is essential for all cell
types, the disease is manifested only in the brain and is
characterized by progressive loss of CNS myelin which
deteriorates upon stress..
We recently developed a knock-in (KI) mice model for the
disease. Our MRI experiments indicated that the mutation
leads to delayed development as well as to neurodegeneration
of specific brain regions, suggesting an important role for
translational regulation in myelin formation and
maintenance.
Various cellular and molecular abnormalities were indicated
in primary cultures of brain cells from the eIF2B-KI mice.

abstract:
Odor perception - discrimination and recognition of volatile
chemicals in the environment – is critical for wide ranging
behaviors including kin recognition, mate selection,
predator avoidance, homing, and feeding. Most naturally
occurring odors are complex mixtures, often containing
hundreds of different components. Furthermore, natural odors
invariably occur against odorous backgrounds. Thus,
olfaction and odor perception involves far more than simple
odor ligands binding to receptors in the nose. I will
describe recent work showing that patterns of neural
activity evoked by complex odors are synthesized into odor
objects by experience-dependent plasticity within olfactory
cortical networks. Furthermore, other forms of cortical
plasticity promote filtering of background odors, allowing
us to smell the roses despite the garbage nearby |

Abstract:
Thermal responses are learned entities, yet very little is
known about the molecules involved in the changes in the
network wiring that underlies this process. In the seminar I
will describe the molecular correlates underlying thermal
control establishment and memory in the hypothalamus.
Furthermore, the epigenetic regulation mechanism which
determines the repertoire of proteins that are involved will
be described concentrating on both the effects of histone
modifications and DNA methylation on the expression of BDNF.

abstract:
Accumulation of cerebral amyloid-β peptide (Aβ) is critical
for developing synaptic and cognitive deficits in
Alzheimer’s disease (AD). However, physiological Aβ
functions, as well as primary mechanisms initiating early
Aβ-mediated synaptic dysfunction, remain largely unknown.
Here we examined the effects of endogenously released Aβ
peptides on synaptic transfer at single presynaptic
terminals and synaptic connections in non-transgenic
hippocampal neurons. Increasing extracellular Aβ
concentration by inhibiting thiorphan-sensitive Aβ-degrading
peptidases rapidly enhanced release probability in
excitatory synaptic connections. This presynaptic
enhancement boosted ongoing neuronal activity and promoted
temporal redistribution of synaptic weights within complex
spike bursts. Notably, both elevation and reduction in Aβ
levels attenuated short-term synaptic facilitation by
bursts. These findings suggest that endogenous Aβ molecules
critically influence regulation of information content
transmitted between neurons. Reduction in synapse capacity
for burst transfer, triggered by Aβ-degradation deficit,
might be a primary pathological event initiating
compensatory synapse loss and memory decline in AD. |
